چکیده

I NTRACELLULAR PRECIPITATES in ghosts prepared from red cells of patients with thalassemia major were described first by Hoffman1 in 1956. Such inclusions have been recognized in intact thalassemic erythrocytes by J osephson et al.,2 Fessas, ’ Nathan et al.46 and others. The present work was stimulated by observations of the difficulty involved in removing hemoglobin from thalassemic ghosts encountered during the course of other studies.7 The work was undertaken to quantitate the residual hemoglobin in ghosts from thalassemic cells in comparison with other hemolytic anemias and normal cells, as well as to compare ghosts from splenectomized versus non-splenectomized thalassemic patients. The role of the spleen in fragmentation of thalassemic cells has also been examined in relation to these intracellular precipitates.

Nonrandom association of free iron with membranes of sickle and beta-thalassemic erythrocytes.

To further define the nature of abnormal iron deposits on the membranes of pathologic red blood cells, we have used sickle cell anemia (HbSS), HbSC, and beta-thalassemic erythrocytes (RBCs) to prepare inside-out membranes (IOM) and insoluble membrane aggregates (AGGs) containing coclustered hemichrome and band 3. Study of IOM from HbSC and thalassemic patients showed that amounts of heme iron a...

Influence of hemoglobin precipitation on erythrocyte metabolism in alpha and beta thalassemia.

Certain aspects of the metabolism of centrifuged young and old erythrocytes in hemoglobin H disease have been examined and compared with similar studies of beta thalassemia and normal cells. Glycolysis, hexose monophosphate shunt activity (HMPS), potassium flux, and glutathione (GSH) content were measured. The distributions of hemoglobins H and F, as well as the activities of erythrocyte glucos...

Cellular and membrane properties of alpha and beta thalassemic erythrocytes are different: implication for differences in clinical manifestations.

To define how excess unpaired alpha- and beta-globin chains in severe beta-thalassemia and severe alpha-thalassemia interacting with the membrane might alter cellular and membrane properties, we performed a series of biophysical and biochemical analyses on erythrocytes obtained from affected patients. Detailed analysis of cellular and membrane deformability characteristics showed that both form...

Intraerythrocytic crystalloid bodies in cats.

SEVERAL HEMOGLOBINOPATHIES of man are known to produce striking morphologic changes in erythrocytes. In vivo sickling, occurring under low oxygen tension, is well documented in hemoglobin 5 (1-lb S) disease. Osmotic dehydration produced by incubation in hypertonic salt solutions fosters sickling in these cells.’ This phenomenon is explained on the basis of a single amino acid substitution, vali...
